Pre-warm the block. Pre-warm the 96-well block to 4 °C (or the protocol's initial hold) before loading the strips. A cold block on a warm strip causes micro-condensation under the cap, which skews qPCR baselines by 0.3–0.8 Ct on the first few cycles.

Cap orientation. Cap the strip in one motion — press the cap strip down flat across all eight tubes in a single sweep, not one tube at a time. A mis-seated cap on even one tube leaks at 95 °C and ruins the run.

Working volume. Keep the reaction volume between 10 µL and 50 µL per tube. Below 10 µL, evaporation losses dominate; above 50 µL, the meniscus sits above the optical read window and Ct values drift upward.

Pipette tip compatibility. Use filtered, low-retention tips for low-copy targets. Standard tips carry over 1–2 % of the master mix, which is enough to fail a single-copy assay.

Seal inspection. After capping, run a fingernail across the cap line. Any tube that gives under the nail is under-seated and has to be re-capped before the run starts. This is the single most common cause of an evaporated column on a CFX96.

Storage after opening. Once the 120-strip pack is opened, store the remainder in the original resealable pouch with the desiccant at room temperature, away from UV. The low-bind surface treatment is sensitive to prolonged UV exposure and to organic-solvent contamination — do not store strips next to a TBE / TAE buffer bath.
